Many times, ductwork runs have anternal dampers installed, so you can 
balance the airflow throughout the house. Find the duct run for that room 
and look for a metal lever sticking out of the bottom of the (usually 
round) supply duct. The damper is open when the lever is aligned with the 
duct, close when it's at 90 degrees.

If that doesn't do it, take the register off and make sure the mechanism 
works properle. Sometimes the mechanism breaks and the register is closed 
even though it looks open.

Finally, check to make sure nothing has blocked the duct at the register. 
My kids are notorious for losing toys down them!
